Generally speaking, and assuming you don't want to bother with additional WAP's, unless you have thick concrete floors the best location for a wifi router is upstairs near the middle of the house. The signal thru wooden floors works pretty well unless there's a mass of cabling or metal piping in there.
WiFi routers are limited to the strength of their signal output, though I can't remember now exactly what it is. I do know you can (legally) buy slightly longer aerials for them which helps a little.
