Am going to replace the grub screw from my front calliper with a speed bleeder valve. Assuming when I remove the grub screw it'll pish brake fluid everywhere so presumably I have to drain it down completely first. And if so am I best pushing in the callipers first?

You'll lose very little brake fluid if you have the replacement bleed nipple at hand ready to insert as you remove the old bleed unit.
